(1) Deep Learning-Driven Development of KEAP1-NRF2 PPI Inhibitors
Summary
My project is to design Deep Learning-based peptide binders that inhibit the KEAP1–NRF2 protein–protein interaction. NRF2 is a transcription factor that switches on genes protecting the cell from oxidative stress, and KEAP1 is the sensor that holds it in check by continuously marking it for degradation. Under normal conditions this balance works well. However, in aging and neurodegenerative disease, oxidative damage accumulates while the NRF2 response becomes blunted, leaving cells unable to mount the protection they need. By blocking the KEAP1–NRF2 interaction, NRF2 escapes degradation and this defense system is reactivated. To achieve this, I use deep learning-based structure design to map the key contact points on KEAP1 and generate peptide binders that engage them selectively.
