(1) Development of a Selective Peptide Inhibitor Targeting the DYRK1A–LIN52 Interaction
Summary
My project explores the development of AI-designed peptide inhibitors that selectively interfere with LIN52 phosphorylation by DYRK1A. This phosphorylation event promotes the assembly of the DREAM complex, a transcriptional regulator associated with cell-cycle control and DNA repair. To determine where selective inhibition may be achieved, I investigate the structural relationship between DYRK1A and LIN52 and search for binding regions suitable for peptide targeting. The identified structural information will guide the generation and optimization of cyclic peptide candidates using protein design tools such as RFdiffusion and ProteinMPNN. Their binding properties and structural stability will first be examined computationally, followed by biochemical testing of promising candidates. Through this work, I hope to develop a peptide-based method for controlling DREAM complex assembly by specifically disrupting DYRK1A-dependent phosphorylation of LIN52.
