(1) AI-Driven Development of Cyclic Peptide Senomorphics Targeting the NEMO–IKK Interaction
Summary
I am working on the development of AI-designed cyclic peptide inhibitors targeting the interaction between NF-κB Essential Modulator (NEMO) and IκB kinase (IKK) to suppress cellular senescence-associated inflammation. Cellular senescence contributes to tissue aging through the secretion of the senescence-associated secretory phenotype (SASP), which promotes chronic inflammation and propagates senescence to neighboring cells. As the master regulator of SASP gene expression, canonical NF-κB signaling represents a therapeutically validated target for senomorphic intervention. Previously developed linear NBD peptides and their peptidomimetics are limited by poor structural preorganization, proteolytic instability, and insufficient blood–brain barrier permeability. To overcome these limitations, this project focuses on the de novo design of cyclic peptide inhibitors that selectively target the NEMO–IKK protein–protein interaction interface through AI-driven structure-based design.
