(1) Deep Learning-Driven Development of Tau Aggregation and Phosphorylation Inhibitors
Summary
I am currently conducting research under the theme of “AI-based Design of Peptide Inhibitors Targeting Tau Protein Aggregation and Phosphorylation.” This study focuses on developing novel therapeutic candidates that can effectively inhibit hyperphosphorylation and aggregation of tau proteins, two major pathological hallmarks of Alzheimer’s disease (AD). In Alzheimer’s disease, tau proteins inside neurons undergo abnormal hyperphosphorylation, which weakens their binding to microtubules. This leads to the formation of intracellular neurofibrillary tangles, ultimately causing neuronal dysfunction and cell death. To block these pathological processes, my research targets both the kinases responsible for tau phosphorylation (such as GSK-3β and CDK5) and the aggregation-prone regions of the tau protein itself. To achieve this, I am leveraging advanced AI-based protein design platforms like RFdiffusion and ProteinMPNN to develop peptide inhibitors that can effectively bind to tau proteins and tau-associated kinases, thereby disrupting their pathological functions. The designed candidates are first evaluated through molecular dynamics simulations and in silico docking studies to predict their binding affinity and structural stability. Subsequent validation will involve cell-based assays and biochemical analyses to assess their actual therapeutic efficacy.
