(1) IDP inhibitor pipeline based on SLiMs
Summary
I am working on developing a general-purpose cyclic peptide inhibitor platform targeting short linear motif (SLiM)-mediated interactions of intrinsically disordered proteins (IDPs), with the goal of enabling broadly applicable dominant-negative inhibition across disease-relevant hubs such as MYC, p53, and β-catenin. Since direct small-molecule targeting of disordered regions remains limited by unresolved selectivity challenges, this project instead focuses on SLiM-mimetic cyclic peptides designed through AI-based structure generation methods such as RFdiffusion and AfCycDesign. Representative protein–protein interaction interfaces, including β-catenin–TCF4 and MYC–MAX, are used as test cases to validate the platform's design pipeline, supported by computational hotspot analyses such as Flex ddG alanine scanning. Through this work, I aim to contribute to translating structural insight into disordered protein interfaces into a generalizable therapeutic targeting strategy.
