(1) Development of a CADASIL Predictive Diagnostic Mode
Summary
I am focusing on a project that develops a predictive diagnostic model for CADASIL based on structural analysis of the NOTCH3 protein. Using protein structure modeling, variant prediction algorithms, and advanced computational and experimental techniques, we identify and predict structural impacts of disease-associated mutations. In particular, we analyze how Cysteine variants within EGF-like repeat domains affect structural stability and the disulfide bond network. The predicted pathogenicity of each variant is validated through in silico analysis and existing clinical and experimental data, ensuring the reliability of the results. The core of my research is to establish a protein structure–based predictive model capable of evaluating CADASIL onset risk and disease severity from patient-derived NOTCH3 variants. Ultimately, we aim to contribute to early and accurate CADASIL diagnosis and the discovery of therapeutic targets for NOTCH3-related disorders.
